Table of contents: Among the autoimmune diseases a large group of them is related to the presence of serum antibodies against nuclear antigens (i.e. Systemic lupus erythematosus, Sjögren syndrome, Scleroderma e.t.c.). Α number of such anti-nuclear antibodies has been identified and determined in daily routine (ds-DNA, SSA, SSB, Sm, snRNP, Jo-1, Scl70, ACA, Histones). Usually, they are firstly determined by screening methods, where the total quantity of antinuclear antibodies is measured, and secondly by quantitative or qualitative methods for each antinuclear antibody separately. In most of cases the indirect fluorescence (IFA) on Hep-2 cells is selected as screening method. IFA results in different images depending of the kind and the location of antigens (homogeneous, speckled, nucleolus, centromere, cytoplasmatic, nuclear rim). Because of the difficulties in daily determination and training of the IFA methods, many laboratories determine antinuclear antibodies with immunoenzymatic methods (ELISA) too. In this case the laboratories find many deviations between the two methods. These deviations are due to the different analytical sensitivity and specifically of the various methods and commercial reagents used and also to the influence of human factor.
